Title: Anther Culture of Fragaria × Ananassa: Environmental Factors and Medium Components Affecting Microspore Divisions and Callus Production

Abstract:
Various environmental factors and medium components were evaluated for their effects on microscope division and callus production in strawberry anther culture. Of the growth regulators investigated, α-naphthaleneacetic acid combined with kinetin produced the highest frequency of calluses. Addition of L-glutamine to the basal medium increased frequency of microspore divisions and callus production in two out of three cultivars tested. Gelling the medium with Gelrite or Gelrite and wheat starch improved callus production. Activated charcoal included in the solid layer, according to the double-layer method, did not affect microspore divisions but inhibited callus growth totally. Low light intensities
during incubation increased callus production as well as the subsequent regeneration frequency. Cold pretreatment increased the frequency of microspore divisions but reduced callus production in two out of three cultivars tested. Regeneration frequency was improved by substituting 6-benzylaminopurine against N-phenyl-N'1,2,3-thiadiazol-5-ylurea. Out of 115 clones so far analyzed, 92 were found to be octaploid (8×) two heptaploid (7×), seven hexadecaploid (16×) and four probably mixoploid (4×–8×).
