Determination of a DNA marker linked to sex in Simarouba glauca DC, an oil-containing tree

Abstract:
Simarouba glauca DC is an economically important tree belonging to the family Simaroubaceae. The tree is cultivated for its edible oil, an ingredient of the human diet for energy and nutrition. It is a dioecious perennial plant with a 4 – 6 year juvenile stage; hence, there is no means to identify the sex of a seedling at the time of planting. Studies were therefore initiated to determine useful sex-linked RAPD markers in Simarouba. Seventy 10-mer Operon primers were selected and used to generate consistent, clear amplification products ranging in size from 250 bp to 4.0 kbp. The primer OPD–20 amplified a band of approx. 900 bp which was consistently present in all female individuals tested. The results showed that RAPD analysis is an effective marker technology with which to develop sex-linked markers to enable the elimination of male seedlings.
